The pathological features of Parkinsons disease are the selective loss of dopaminergic neurons in the substantia nigra pars compacta (SNpc),and the presence of Lewy bodies (LB) which contain alpha-synuclein (αSNY) in the surviving SNpc dopaminergic neurons.Triplication of the wild-type (WT) locus has been linked to autosomal dominated Parkinson s.Recombinant adeno-associated viral (rAAV) vector-mediated overexpression of αSNY has been shown to cause neurodegeneration of the nigrostriatal dopaminergic pathway in rodents and primates. But we do not know the exact physiological and pathological feature of αSNY.The subcellular map of αSNY in rat SNpc neurons will provide new insight into these questions. In this work, the rAAV carrying αSNY cDNA (rAAV-oαSNY),and rAAV were unilaterally injected into the SNpc.
