Aim of study:Insulin resistance is the "common soil" of diabetes and cardiovascular disease.With significant morbidity,its targeted therapy method is still absent at present.Our Previous study shows that Fufang Zhenzhu Tiaozhi Fang(FTZ) which is a approved Chinese herbal prescription with unambiguous effect on dyslipidemia can improve insulin resistant in vitro.This study conducted a further investigate on the insulin sencitivity of FTZ in vivo.As we known,Adipose tissue is the largest endocrine organ which is closely related with insulin resistance,whether it involved in the beneficial effect of FTZ was also detected.Material and methods:Male wildtype aP2 mice (aP2-W mice) and C57 mice was giving a high-fat feeding for 5 months as well as FTZ intragastric administration.Food intake,body weight was measured while blood glucose and plasma triglyceride were determined.Insulin sensitivity was evaluated by an oral glucose tolerance test (OGTT).We used aP2 diabetic transgene mice (aP2-T mice) which lack of mature adipose tissue as a model to detect the role of adipose tissue in insulin sensitivity regulation by FTZ.AP2-T mice accept the same FTZ intake,and use Arabic gum as control.After 5-month treatment,all the mice was sacrificed,hepatic and muscular lipid content were determined.Results:FTZ giving resulted a lower blood glucose and plasma triglyceride level in both wildtype and transgene aP2 mice.The area under curve of OGTT was significantly decreased in high fat feeding aP2-W mice,the same result was obtained in C57 mice but not the lipoatrophic aP2-T mice.VVhats more,the muscular lipid content was obviously lower in aP2-W mice although there were not significant change in hepatic lipid content.Conclusions:FTZ can improve high fat diet-induced insulin resistance,and its mechanism can be related with adipose tissue.
