目的:探讨Meis1在围着床期小鼠子宫内膜中的作用。方法:采用免疫组织化学定位检测Meis1在小鼠子宫内膜中的表达;采用Real-time RT-PCR和免疫印迹方法,分别在mRNA和蛋白水平定量检测Meis1在围着床期小鼠子宫内膜中的表达水平。结果:免疫组织化学结果显示,Meis1蛋白主要表达于小鼠子宫内膜腺上皮细胞的细胞膜和胞质以及基质细胞和蜕膜细胞的细胞核中,在围着床期小鼠子宫内膜中的表达随妊娠天数的增加而增加,着床后在基质和蜕膜中的表达增强;在孕第4日显著增加,孕第5日达高峰,孕第6日表达下降。Real-time RT-PCR和免疫印迹结果显示,Meis1的表达随妊娠天数的增加逐渐增强,孕第4日明显增强,孕第5日达高峰,孕第6日开始下降。结论:Meis1在围着床期小鼠子宫内膜中呈规律表达,与小鼠着床窗吻合,提示Meis1是调控围着床期子宫内膜容受性的重要因子。
Objective: To investigate the role of Meis1 in the peri-implantation mouse endometrium. Methods: The expression of Meis1 was detected by immunohistochemistry in mouse endometrium. Real-time RT-PCR and Western blotting were used to detect the expression of Meis1 in the perinatal mouse endometrium In the expression level. Results: Immunohistochemical results showed that Meis1 protein was mainly expressed in the cell membrane and cytoplasm of mouse endometrial glandular epithelial cells and in the nuclei of stromal cells and decidual cells, and was expressed in peri-implantation mouse endometrium Increased with the increase of the number of gestational days, increased in the matrix and decidua after implantation, significantly increased on the 4th day of pregnancy, reached the peak on the 5th day of pregnancy and decreased on the 6th day of pregnancy. The results of Real-time RT-PCR and Western blotting showed that the expression of Meis1 gradually increased with the number of days of pregnancy, and significantly increased on the 4th day of pregnancy. It reached the peak on the 5th day of pregnancy and began to decrease on the 6th day of pregnancy. Conclusion: Meis1 is expressed regularly in peri-implantation mouse endometrium and coincides with mouse implantation window, suggesting that Meis1 plays an important role in regulating endometrial receptivity.
