低温胁迫下菊花蛋白质组的双向电泳分析与质谱鉴定

  通过双向电泳(2-D electrophoresis)和基质辅助激光解析电离飞行时间质谱技术(matrix assisted laser desomtion/ionization time of flight lnass spetrometry,MALDI-TOF-MS)对抗寒性不同的菊花品种进行了蛋白质组学的比较分析.以敏寒品种北京小菊(Dendranthema cv.pekinense)和耐寒品种地被菊美矮黄(Dendranthema×grandiflorumMeiaihuang)为材料,4℃低温处理0 hr、2 hr、4 hr、12 hr、24 hr、48 hr、72 hr 后分别提取叶片总蛋白,采用pH3-10 线性IPG干胶条进行双向电泳分离,染色后利用Image Maste-2D Elite 图象分析软件进行处理;通过斑点配比,获得了不同时间低温处理下北京小菊差异蛋白54 个,地被菊美矮黄差异蛋白52 个,以及这两个菊花品种在相同低温处理时间下的差异蛋白7 个;分别对这些差异蛋白采用MALDI-TOF-MS 进行肽质谱指纹图谱(Peptide Mass Fingerprinting,PMF)分析,共获得了63 个较好的肽质量指纹图谱,通过在线MASCOT 软件进行搜索比对,初步鉴定了12 个差异蛋白,并分析了各自功能及与低温的关系,探讨性地提出了菊花的抗寒机制.
