研究背景:调节性T细胞(Treg)在建立和维持外周免疫耐受中具有十分重要的作用.Ⅰ型调节性T细胞(Tr1)是初始CD4+T细胞在外周组织中存在IL-10的条件下,遇到外来或组织特异性自身抗原时被诱导产生的,不表达Foxp3,主要通过其进一步分泌抑制性细胞因子IL-10发挥免疫调节功能.我们以往的研究证实,无论是APC表面的膜型B7-H1分子,还是可溶性B7H1-Ig融合蛋白,在体外均能作为共刺激分子,诱导小鼠CD4+T细胞分化为IL-10hhighIL-5highIFN-γmediumIL-4lowIL-2low/Foxp3-的Tr1细胞.许多报道显示,细胞因子IL-21与IL-10的产生密切相关.
