Objective: Activation of transient receptor potential vanilloid subtype 1 (TRPV1) increases paracellular permeability of submandibular gland, but its underlying mechanism is unknown.Occludin is the first identified transmembrane tight junction protein, which acts as a main functional molecule in forming the tight junction barrier in a variety of tissues and cells.The aim of this study was to investigate the effect of TRPV 1 activation on occludin properties and the possible signaling pathway.Methods: The expression of TRPV1 and occludin were detected by RT-PCR and Western blot.The distribution of TRPV1, occludin, claudin-3,-4, E-cadherin, β-catenin, and filamentous actin (F-actin) were observed by immunofluorescence.The effect of capsaicin on tight junction function was evaluated by transepithelial electrical resistance (TER) measurement and paracellular permeability assay.The role of occludin was investigated by transfection with either occludin shRNA or cDNA.Phosphorylation of extracellular signal-regulated kinase (ERK) and myosin light chain 2 (MLC2) were detected by Western blot.
