  Cherriistry and Molecular Sciences,Wuhan University Wuhan 430072,PR China Foodborne illnesses have always been a serious problem that threats public health,so it is urgent to develop a method that can rapidly and sensitively detect pathogens at low levels in food.Immunomagnetic isolation in microfluidics has emerged as a technique for the separation and enrichment of pathogens.In this paper,we reported a magnetic-controlled microfluidic system [l] which integrated the separation,enrichment and detection of S.typhimurium.The antibody modified magnetic nanospheres synthesized by LBL method [2] can specifically capture the S.typhimurium.Then the magnetically captured S.typhimurium were separated,enriched and detected in a single chip under a fluorescence microscopy based on the principle of sandwich immunoassays.In addition,this method had excellent selectivity and specificity,good reproducibility and strong anti-interference ability.It had been successfully used to detect S.typhimurium synthetic samples(milk,fetal bovine serum).The developed system provides a rapid and sensitive tool to image and quantify pathogens at low levels with large sample volumes in practice.
