[Objective] The study aims to construct a recombinant adenovirus vector containing FAM3c gene, evaluate the transfection efficiency and identify its function in mouse glomerular mesangial cells (Mcs).[Methods] FAM3c gene was amplified by PCR from mouse kidney cDNA library, and inserted into pEASY-1 vector using T3 ligase.Single scissored by Bgl Ⅱ, FAM3c gene was inserted into pAdTrack-CMV with T4 ligase.Linearized by PmeI and electronically transfected into BJ5183, the Ad-FAM3c was then packaged by transfecting into HEK293 cells and identified by infectiing glomerular mesangial cells (MMcs) of mouse.Injected Ad-FAM3c into the mouse attacked by mesangial proliferative glomerul onephritis, the cells of the mouse were observed by HE staining.[Resultsl DNA sequencing and agarose gel electrophoresis pictures proved FAM3c was inserted into the vectors.On the picture of western blot the expression of Ad-FAM3c was enhanced in MMcs.Pathological sections indicated Ad-FAM3c increase inflammatory response.[Conclusion] Ad-FAM3c was successfully constructed.FAM3c gene plays an important role in mesangial proliferative glomerul onephritis.
