DYNAMICS OF DNA HYBRIDAZATION STUDIED USING A MICROFLUIDIC MULTI CHANNEL QUARTZ CRYSTAL MICROBALANCE

  This paper describes a novel system to study hybridization dynamics of DNA by using a quartz crystal microbalance (QCM) as transducer and microfluidic chip (MFC) as the processor,which is constructed of a MFC,a multipal channel QCM chip and connecting device between them.In this system an automatic control device,which could automatically control the multiple time consuming procedures,such as ss-DNA fixation,wash,dry process,target-DNA hybridization etc.,was developed,greatly saving the time and enhancing the accuracy of the QCM measurements as compared with those by manual operation in the market.Take the advantage of this innovation we can do many complicated ss-DNA fixation and target-DNA hybridasation (2nd layer adsorption) events with a small amount of samples in a relative short time.We demonstrate and validate this fact by using a fixation and hybridization experiment of a pair of DNA coming from P53 gene,single strain DNA Oligol and target DNA Oligo2.By means of the enhancement of 12nm-Au-nanoparticles made in the MFC,detection sensitivity of Oliger 2 higher than 10-13mol/L was obtained.The time for an Oligol fixation kinetic curve construction as well as the time for the hybridization of Oligol and Oligo2 only takes about several dozens of min.We can directly get the apparent combination coefficient Kcom of Oligo1 and Oligo2 according to a Langrnuir adsorption isotherms from the obtained hybridization isotherm.The parameters of all data in this system could be real time controlled and obtained with an ordinary computer.In addition to it,connection of MFC to QCM could make the samples more pure and concentrated; amplification of sensitivity by nanoparticle technology can further improve the selectivity as well as the accuracy of measurement.
