Enantioselectivity of o,p′-DDT and its Influence on MCF-7 Breast Cancer Cell Migration and Infiltrat

  The great concern over DDT exposure and breast cancer has been aroused for few decades.With a chiral center, o,p′-DDT exerts the strongest estrogen effect among DDTs, and R-o,p′-DDT is more potent than S-o,p′-DDT.However, the enantioselectivity impact on occurrence and development of breast cancer of o,p′-DDT still remains to be elucidated.In the study, o,p′-DDT could enantioselectively interfere breast cells migration and infiltration and related genes expression were evaluated in vitro MCF-7 cells.Both cells migration and infiltration were induced by o,p′-DDT,and R-o,p′-DDT had greater effect than S-o,p′-DDT.Furthermore,the mRNA expression of MMP-2,9 and hTERT were upregulated by R-o,p′-DDT exposure.Meanwhile the R-o,p′-DDT significantly inhibited the mRNA expression of TIMP-1,and TIMP-4 and cell adhesion molecules.For our knowledge,this is the first report about chiral estrogenic chemicals enantioselective impact on the development of breast cancer.Our results suggest that the estrogenic like chiral compounds are of significant concern with respect to human cancers depended by estrogen.
