Purpose: To prepare well-designed cationic proliposomes-mediated asODN delivery system;Methods:A thin film-rehydration-lyophilization method was developed.The morphology was investigated by transmission electron microscope(TEM).The diameters and surface charge were determined by laser particle analyter.Dissociated asODN were disparted from the liposomes by sephadex G-50 column and entrapment efficiency (EE%) were measured by UV detection at 260nm.The stability was studied by long-term storage at 4℃; Results: The proliposomes possess excellent stability during long-term storage at 4℃ and rapid redispersion with trehalose as lyoprotectant.The diameters were 100~150nm with EE% of 88.4% pre-and 150~200nm with EE% of 83.2% post-lyophilization respectively.Mean zeta potentials were + 45mv; Conclusion: The method was successful.
