Human Lef-1 stop codon readthrough produces a functional extended isoform and regulates Siglec-5 exp

  Lef-1 is a terminal effector in the wnt signaling pathway.There are several Lef-1 isoforms derived from either pre-RNA splicing or alternative promoter usage.We have identified a Lef-1 isoform formed via UGA stop codon readthrough.This full length Lef-1 isoform is identical to Lef-1 isoform-2 from chimpanzee, which has no in-frame premature stop codon.3D-structure analysis shows an extra HMG-BOX and two nuclear receptor-Boxes in extended peptide.In vitro transfection studies show the extended peptide can enhance HIV tat activation effects on the HIV-LTR;however, the long fusion readthrough isoform can reduce HIV-LTR activation.Stop codon readthrough of Lef-I can be enhanced by some aminoglycosides.
