Canine parvovirus type 2 (CPV-2) causes a highly contagious and often fatal disease is characterized by vomiting and haemorrhagic gastroenteritis in dogs of all age and myocarditis in pups of less than 3 months of age.Active virus circulation and initial vaccination programmes helped develop herd immunity in canine populations, which greatly reduced mortality and further spreading of the virus. However, host-immunity pressure may have also contributed to the progressive emergence of CPV-2 antigenic variants. At the past decades, new antigenic types of the CPV-2, CPV-2a, CPV-2b and CPV-2c, were frequently reported in the dog population and replaced the original CPV-2, which has caused widespread concern. The VP2 of the CPV-2 is the major capsid protein, and it is highly antigenic, playing important roles in determining viral host ranges and tissue tropisms. The CPV-2 strains could be assigned to one CPV-2 type based in the presence of a deduced asparagine (2a), aspartic acid (2b) or a glutamic acid (2c) in the amino acid position 426 of the VP2 protein. At present, the VP2 gene was widely used for identification and monitoring of the types of CPV-2 circulating in the canine population. In the present study, a total of 201 stool samples were collected from dogs with diarrhea in Heilongjiang Province of northeast China from May 2014 to April 2015, and were analyzed by polymerase chain reaction (PCR) to partially amplify for the VP2 gene (583 bp) of CPV-2.Results revealed that 95 samples (47.26%) were positive for CPV-2 and showed 98.8% -100% nucleotide identity and 97.6%-100% amino acid identity;of 95 positive samples, the types new2a, new2b and 2c accounted for 64.21% (61/95), 21.05% (20/95) and 14.74% (14/95), respectively.Resulting data revealed that the current dominant viral variant in northeast China is new CPV-2a, but new CPV-2b and CPV-2c were found in fecal samples of diarrhea-affected dog.
