帕金森病致病基因DJ-1相互作用蛋白蛋白质组学及线粒体功能调节研究

  目的 帕金森病是一种常见的神经系统退行性疾病,其具体致病机制尚不明确.J-1是常染色体隐性遗传性PD PARK7型的致病基因,该基因突变导致的PD具有起病早和临床表现典型的特征.由于DJ-1具有抗氧化等多种功能,所以DJ-1在帕金森病中所发挥的重要作用受到人们越来越多的关注.我们应用蛋白质组学的方法通过寻找DJ-1相互作用蛋白来探讨其具体生理功能及致病机制.方法 采用GST Pull down偶联质谱鉴定的方法,结合生物信息学分析以及体内外研究,寻找DJ-1的相互作用蛋白及其参与的生物学功能和通路.揭示DJ-1的生物调节功能及其所参与的帕金森病致病机制.结果 利用原核表达以及免疫亲和吸附方法获得高浓度、高纯度的DJ-1蛋白.通过Pull down偶联质谱鉴定技术分离鉴定出441个DJ-1相互作用蛋白.应用生物信息学分析方法注释、分类DJ-1相互作用蛋白.结果表明DJ-1在体内与多个蛋白相互作用,这些相互作用蛋白分布于胞浆、细胞核以及各种细胞器;涉及氧化应激、能量代谢、生长增殖、免疫调控等多个生物过程,调节MAPK、mTOR、VEGF等多条信号通路;参与阿尔兹海默病、帕金森病、乳腺癌、黑色素细胞瘤等多种疾病;此外,体外细胞模型中我们发现D J-1与DNM1L在细胞内存在良好的共定位,在功能上存在相互作用关系.DJ-1通过调节DNM1L的磷酸化修饰水平,发挥线粒体形态功能调节作用,导致帕金森病发病.结论 DJ-1参与多个生物调节过程以及致病机制.DJ-1通过与DNM1L的相互作用调节线粒体形态功能参与帕金森病发病.
