Terpenoids are a class of compound isoprene as a structural unit.More than 30,000 naturally occurring terpenoids have been reported,and the number is growing steadily.Terpenoids have a variety of biological functions and are widely used in pharmaceutical,food,chemical,etc.By getting the benefits of metabolic engineering and synthetic biology approaches,heterogeneous production by microbial cell factory is becoming a cheap and promising way to produce terpenoids.Terpenoids are biosynthesized from only two precursors: isopentenyl pyrophosphate(IPP)and dimethylallyl pyrophosphate(DMAPP).The level of IPP&DMAPP is a key factor to get the high-yield production of terpenoids.In metabolic engineering study,it can be indicated by the color change of lycopene production.Lycopene is a linear carotenoid composed of six isopentenyl diphosphate(IPP)and two dimethylallyl diphosphate(DMAPP).As an effective antioxidant,it has been proposed as a possible treatment for some cancers.Thus lycopene biosynthesis module will not only be important for lycopene production,but also provide a tool to study terpenoids pathways.Here we investigated carotenoid genes(crtE,crtB,and crtI)from diverse sources.A series of lycopene biosynthesis modules were then constructed.These modules will be helpful for both MVA and MEP pathway studies.
