负性协同刺激分子B7-H4在C3H10T1/2移植治疗EAE中作用机制的研究

  目的:探讨负性协同刺激分子B7-H4在C3H10T1/2细胞移植治疗小鼠EAE模型的体内效应机制。方法:利用MOG35-55和完全弗氏佐剂制备小鼠实验性自身免疫性脑脊髓炎(EAE)模型,将n=50只小鼠分为正常对照组、EAE组、C3H10组(移植C3H10细胞)、C3H10-NC组(移植shRNA对照质粒转染的C3H10细胞)、C3H10-B7-H4组(移植B7-H4靶向shRNA转染的C3H10细胞),之后每日对各组小鼠进行神经功能缺损评分,在免疫后第6天,按1×106细胞每只小鼠分别移植入小鼠体内;在小鼠发病的急性期,用流式细胞仪检测各组小鼠外周血细胞上B7-H4的表达变化;用ELISA法检测外周血细胞因子的表达变化;收集小鼠脾脏组织,流式细胞仪检测各组脾脏细胞上B7-H4的表达变化;结合神经功能评分观察不同十预方式对EAE损伤组织结构和功能的影响,明确B7-H4分子在C3H10移植治疗EAE体内效应的机制。
