Adipose tissue inflammation is the key linking obesity to insulin resistance.Over 50% of the interstitial cells in adipose tissue are macrophages (adipose tissue macrophages, ATMs), which produce inflammatory cytokines and therefore play an important role in the progression of insulin resistance.Two types of macrophage have been defined: M1 (proinflammatory) and M2 (anti-inflammatory).Des-acyl ghrelin, which was originally reported as the inactive form of ghrelin and accounts for more than 90% of total circulating ghrelin, could increase insulin sensitivity, but the mechanism is still unclear.Here, we reported that administration of des-acyl ghrelin to mice inhibits the inflammation of adipose tissue by regulating the ratio of M 1 and M2.In vitro study also showed that ATMs undergo a phenotypic switch from M1 to M2 upon des-acyl ghrelin stimulation.Thus, des-acyl ghrelin induces a phenotypic switch in ATMs polarization and subsequently increases insulin sensitivity.
