The study focuses on developing Chinese medicine granules as Western medicine pellets using a fluid-bed rotor granulator; combined with a factorial experimental design in the experiments.Tile characteristc adventure of the traditional Chinese medicine granules is retained such as easily disintegrated characteristic.Moreover, process to make agglomerized, so called spheronization, was conducted in order to improve the flowability, reduce friability and prolong shelf life as well.To evaluate the effects on the produced pellets and dependent variables by employing a 2n-1 factorial experimental design, four factors that influence the production are set as: (1) ratio of excipients (2) inlet temperature (3) spray rate (4) rotor speed.Furthermore, we evaluate other physical properties of pellets such as content of TanshinoneIIA and Salvianolic acid, angle of repose, friability, particle size distribution, geometric mean diameter, geometric standard deviation and disintegration time.Finally we observe the pellet weight change due to water absorbed in RH 75% at 40℃ condition so that we are able to assess the stability of shelf life.The experiments indicate that TanshinoneIIA, Salvianolic acid, the marker compounds, are also kept constant.Meanwhile we identify the marker compotmds are rarely destroyed under experimental variables and provide content uniformity too.Two dependent variables are analyzed by statistical evaluation, it showed that the square root of drug loading were affected by the four independents, and likewise the paired interactions such as excipient ratio and inlet temperature, excipients ratio and spray rate.Moreover, the four independents, as well as the interactions among the variables related to excipient ratio, had effects on the square root of geometric mean diameter.Besides, pellets are more advantaged than those traditional granules by their smaller angle of repose, reduced friability, and required legislative standards of disintegration time.And, it is noticeable that pellets feature anti-rewetting at high relative humidity.This helps extend shelf-life of the products and promote Chinese herbal preparations in both quality and quantity.
