Objective Literatures have shown that strong associations between HLA-B* 1502 and Stevens Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) induced by carbamazepine in Hong Kong and Guangzhou areas.There are some conflicts between primers in the papers.The aim of this project is to find correct methods to determine HLA-B*1502.Methods We enrolled 138 cases including the SJS/TEN patients, milder non-SJS/TEN cutaneous adverse drug reactions (mainly maculopapular) patients, tolerant controls and normal subjects.PCR-SSP was performed using sets of HLA-specific primers and control primers to genotype HLA-B* 1502 and high-resolution genotyping was used to sequence HAL-B* 1502.Results After testing primers of published papers, we found that 6 cases should be HLA-B* 1502 carriers.But among them, high-resolution genotyping proved that only one was HLA-B* 1502 carrier.We analyzed HLA-B* 1502 sequence and designed new primers for genotyping HLA-B*1502 and confirmed that testing results were exact same with sequence results.Conclusion New primers used to genotype HLA-B* 1502 have been found.These strategies can help to guide personalized medicine and predict severe cutaneous adverse reaction in the clinic.
