To better understand the expression patterns of Gnαq in formation of mammalian coat color, mRNA expression was detected by real-time fluorescent quantitative polymerase chain reaction (PCR);cellular localization of Gnaq in white and black skins were examined by immunohistochemistry;and qualitative investigation of Gnαq protein was analyzed by Western blot.The results showed high homology of sheep Gnαq cDNA sequence (98 %).The result was corrected by sequence analysis.In white and black sheep skins, Gnαq protein was localized to hair shaft, inner root sheath, dermal papilla and hair matrix.The amounts of Gnαq mRNA and Gnαq protein in black sheep skins were significantly greater than that of in white sheep skins (P 0.01).In conclusion, Gnαq is expressed in different coat colors.These results suggest that Gnαq is correlated with pigment formation in different coat colors.
