The aim of the study was to investigate the anti-apoptotic effect of an endogenous neuropeptide,ghrelin on the mouse MPTP model of PD.Ghrelin,a stomach-derived hormone inducing growth hormone (GH) release and promotes positive energy balance,has been reported to stimulate proliferation and inhibit apoptosis in several cell types.By immuno-fluorescence staining,we showed that growth hormone secretagogue receptor (GHS-R) 1 a expressed on dopamine neurons in the SNpc in mice.Further studies by HPLC and immunohistochemistry showed that ghrelin could protect against MPTP-induced dopamine depletion in the striatum as well as the dopaminergic neuronal loss in the Substatia nigra.The RT-PCR studies demonstrated that pre-treatment with ghrelin inhibited MPTP-induced down-regulation of caspase-3,Bcl-2 mRNA expression,and up-regulation of Bax mRNA expression in the SNpc.These effects were fully abolished by the antagonist for GHS-R1a,D-Lys3-GHRP-6.This study is the first to demonstrate that the neuroprotective effect of ghrelin on dopaminergic neurons in MPTP-treated PD mouse model was through the anti-apoptotic pathway.
