Drosophila katanin functions at neuromuscular synapses and is required for microtubule network forma

  Katanin is a member of the highly diverse AAA (ATPases associated with different cellular activities) protein superfamily.Katanin, a heterodimer of 60-and 80-kDa subunits (p60 and p80), uses energy from ATP hydrolysis to disrupt tubulin contacts within the microtubule (MT) lattice, severing the MT but leaving tubulin capable of subsequent repolymerization.Dm-Kat60 (Drosophila katanin-60, CG10229) could disrupt MT arrays when overexpressed in S2 cells and play important role in neuronal development.Up to now, in vivo function of katanin have not characterized in model animal except worms.To dissect the functions of katanin, we made katanin-60 null mutant, transgenic UAS and RNAi flies which can be respectively used to over and knockdown the expression of CG10229, which encodes katanin-60, Drosophila ortholog of human katanin-60, a-tubulin stanining of 3rd instar larva muscle of katanin-60 mutants showed that the microtubule network is increased in perinuclear MT density compared to in wildtype.Tissue specific overexpression of katanin-60 in muscle resulted in damage in MT networks around nuclears at 25 degree and totally disrupted MT massworks at 28 degree respectively.Alterations of katanin-60 expression also affected neuromuscular synapses.
