溶菌酶热变性机理的拉曼光谱研究

小蛋白的变性机理是人们关心的热点,对一些小蛋白的单步骤和多步骤机理一直存在争议.这里我们利用拉曼光谱研究了溶菌酶的热变性机理.我们首次对所有波段的光谱进行了完整分析,我们发现侧链基团相关的拉曼谱峰在较低温~74摄氏度就发生了转变,但是主链基团相关的拉曼谱峰在较高温度~76.5摄氏度才发生转变,这提供了溶菌酶热变性两步骤的直接证据.通过光谱分析,我们知道侧链基团对蛋白质的三级结构更加敏感,主链基团对二级结构更加敏感,这表明热变形时溶菌酶的三级结构先变得松散,然后水分子进入这个松散的溶菌酶分子进一步诱导二级结构的转变,整个热变性过程中存在着一个亚稳态的中间体.我们的结果不仅直接揭示了溶菌酶的两步骤热变性机理,还表明完整的分析所有拉曼谱段是研究蛋白质变性机理的一个非常有效的方法.
