Type 1 diabetes (T1D)is an autoimmune disease resulting from self-destruction of insulin-producing β-cells.Neutrophils have recently been implicated in both the onset and progression of T1D.Reduced neutrophil counts have been observed in patients with T1D, and the neutrophil infiltration and NET formation in islets can be observed as early as two weeks after birth in non-obese diabetic (NOD) mice. However, how the neutrophils participate in the T1D pathogenesis remains elusive. Our clinical studies demonstrated that augmented NETosis and increased neutrophil elastase (NE) and proteinase 3 (PR3), which are two major members of neutrophil serine proteases (NSPs) stored within azurophil granules of neutrophils, are closely associated with β-cell autoimmunity in patients with T1D, especially those with disease duration of less than 1 year.
