Preparative Isolation and Purification of Two Compounds from Radix Angelicae Sinensis by High-Speed

  In the present study,high-speed counter-current chromatography was applied to the systematic separation and purification of chemical components from the ethyl acetate extraction of Radix Angelicae sinensis extracts.The optimized solvent system,consisting of n-hexane-ethyl acetate-methanol-water [1] (1:2:0.8:1 VNN/V),was performed to isolate and purify the two aim chemical components in using of high-speed counter-current chromatography (HSCCC).Subsequently,the fractions of HSCCC were analyzed by high performance liquid chromatography (HPLC).The purity of the two compounds was 96.8% and 99.9% respectively.And the two compounds were identified by 1HNMR,MS and IR.The results demonstrated that HSCCC is a speedy and efficient technique for systematic isolation components from traditional medicinal herbs.
