Abnormal glycosylation of glycoprotein often associates with development of disease.However, little is known about the relationship between the specific glycopattems of glycoprotein in human body fluids (saliva, serum and urine) and type Ⅱ diabetes.The aim of this study was to investigate the specific glycopatterns of glycoproteins in body fluids of patients with type Ⅱ diabetes and reveal the correlation between the specific glycopatterns in human body fluids and type Ⅱ diabetes.Body fluid samples (whole saliva, serum and urine) were collected from patients and healthy controls.According to the age and genders, the specimens from patients and healthy controls were divided into three groups (young men, elderly men and elderly women).Lectin microarrays were utilized to investigate the glycopatterns of glycoproteins in body fluids of patients and healthy controls.The results revealed that expression level of fucoseα1-2Galβ1-4GlcNAc, Galβ-1,4GlcNAc, αGalNAc and terminal GalNAc were increased, meanwhile, fucoseα-1,6GlcNAc and α fucose were decreased in saliva specimens of all patients groups.Expression level of fucoseα1-2Galβ1-4GlcNAc,terminal GalNAc and Sia2-3Galβ1-4GlcNAc were increased in all specimens of body fluids from young men.Body fluid protein microarrays and lectin blotting were used to verify the terminal carbohydrate moieties of glycoproteins in individual samples.
