The polymorphism of the Beta3-Adrenoceptors gene (ADRB3) has an association with lambs cold survival. To detect the Single Nucleotide Polymorphisms in the ADRB3 gene in Chinese sheep populations, we involved total 421 blood samples from 6 different Chinese local sheep populations (Hu, Small-tail Han, Ganjia, Ola, Qiaoke and Merino crossbred). The polymorphism of a 263bp region in the intron of the ADRB3 gene was analyzed by PCR-SSCP. As the result shows, total 12 novel SSCP patterns are detected. The sequences of these 5 new patterns identified by cloning and sequencing were proved as representatives of new ovine ADRB3 alleles (Genbank reference No. EU371403-EU371407). A deletion and 2 new Single Nucleotide Polymorphisms (SNPs) are deducted which add the number of SNPs in this region from 8 to 10. Furthermore, some of the new alleles were population-special which suggests they have potential link with population-special traits. In future, the association between polymorphism of the ADRB3 gene and cold tolerance therefore will been further confirmed by sheeps cold-survival phenotype investigation in China high plateau region, so that the ADRB3 gene can be employed as a gene marker in lambs cold tolerance testing.
