Linear prenyl pyrophosphates are the key precursors of many important plant terpenoids:limonene, linalool, abscisic acid, gibberellic acid, chlorophyll.Prenyltransferase (PTS)account for the majority biosynthesis of linear prenyl pyrophosphates.Arabidopsis thaliana possess a group of PTSs whose functions are not clearly determined.We tested the activity of ten AtPTSs and classified them into three different groups: GGPS (C20), GFPS (C25),and >C25 PTSs.By determining the representative structures, the underlying mechanism of product chain length determination of these AtPTSs is proposed, and we describe it as a "three floor" model.This model is proved by site-directed mutagenesis and enzymatic analyses.Using this model, we succeed in prediction novel GFPSs in various plant species.
