Production of plant bioreactor is important for the total area of genetically modified crops, which continues to increase in recent years.As the gene transformation receptor of plant bioreactor, oil body system not only improves the expression level of heterologous protein, but also reduces the cost of whose separation and purification.In this study, an efficient construct of plant oil body expression system has been established, which contain a seed-specific promoter (OP), an oleosin gene and the terminator of a seed-specific gene.The gene encoding the human basic fibroblast cell growth factor (bFGF) was then inserted between the oleosin gene and the terminator, which resulted in the construct of fusion gene of Oleosin::bFGF driven by the seed-specific promoter.The fusion gene was successfully transferred into Arabidopsis and the T3 generation homozygous transgenic Arabidopsis lines were obtained.Western-blot and ELISA analysis indicated that bFGF was expressed in transgenic Arabidopsis seeds, whose expression level is 75.6ng/g dry seed.The Oleosin-bFGF fusion protein was purified by gradient centrifugation, and the purified Oleosin-bFGF has showed a significant effect on the proliferation with NIH 3T3 cells.This indicated the Oleosin-bFGF fusion protein via the oil body system in plant was biologically active, which makes it possible to produce the therapeutic proteins by plant bioreactor.
