Alzheimers disease(AD)is a progressive and fatal brain disorder that causes memory loss,steady deterioration of cognition,and dementia.Although the exact cause of the disease is under intense investigation,the prevailing hypothesis proposes that the low levels of acetylcholine(ACh),amyloid β-protein(Aβ)deposits,oxidative stress and neurofibrillary tangles,which seem to play important roles in the disease.However,current treatment of AD focuses on increasing cholinergic neurotransmission in the brain with acetylcholinesterase inhibitors and inhibiting N-methyl-D-aspartate(NMDA)receptor with memantine.But in clinical trials these drugs have been proved for treatment only mild to moderate severity cases,and do not reverse or heal the disease.Due to the multifaceted etiology of AD and unsufficient efficacy of single drug,it is necessary to develop new anti-Alzheimer agents with multiple potencies.
