Objective: Establish and validation a HPLC for determination tacrolimus.Methods: Chromatographic column: Ultimate(R) XB-Diol (4.6 × 250mm, 5 μ m) (Welch Materials,Inc.), Mobile phase: hexane::l-butyl chloride: acetonitrile; Detection Wavelength was 225nm, flow rate: 1.7ml/min; Column temperature: ambient temperature; Results: The tacrolimus linear range was from 2 μ g/ml to 200 μ g/ml (r=0.9999), Linear equation y =2600.7x-583.82, the average recovery was 98.97%, and RSD =0.47%.Conclusion: This method was simple, sensitive and accurate, reproducible and strong specificity, Can be used for tacrolimus ointment of tacrolimus is determined.
