Nanocarriers delivering prodrugs are important strategies to improve the in vivo effectiveness and efficiency.For therapeutical efficacy, the prodrug must hydrolyze to its parent drug after administration.In this study, we present proof-of-concept and methodology in support of our hypothesis that increased steric hindrance and decreased polarity are major handicaps for hydrolysis.
