目的:体外建立心肌微血管内皮细胞(cardiac microvascular endothelial cells,CMVEC)的实验模型,探讨干扰Notch信号通道对心肌微血管内皮细胞间质转分化的作用.方法:采用体外酶消化法分离培养大鼠CMVEC,通过两次差速贴壁纯化细胞.应用倒置相差显微镜观察细胞形态,免疫荧光法检测细胞表面特异性抗原.构建Notch1RNAi慢病毒.实验分为三组:A:正常对照组
