Streptococcus suis serotype 2 (S.suis 2) is an emerging zoonotic pathogen that causedtwo outbreaks of streptococcal toxic shock syndrome (STSS) in China. Our previous study demonstrated that a type V secretion system (T4SS)harbored in the 89K pathogenicity island (PAI) contributes to the pathogenicity of S.suis 2.In this study,we used a shotgun proteomic approach to identify the effectors secreted by the T4SS in S suis 2 and identified surface-associated subtilisin-like protease-1 (SspA-1) as a potential virulence effector. Both western blot analysis and pull-down assayshowedthat secretion of SspA-1 depends on the T4SS.
