  The insulin-like growth factor/IGF/signaling system plays an important role in the regulation of cell proliferation in many tissues.The IGF system is comprised ofIGF ligands,cell surface receptors/IGF-1R and IGF-2R/,and IGF-binding proteins.IGFs stimulate cell proliferation and exert mitogenic and anti-apoptotic functions in tumor cells through IGF-1R,which possesses ligand-activated tyrosine kinase activity.It was recently shown that IGF-1R was overexpressed in number of cancers,including non-small cell lung cancer.A promising strategy to inhibit the function of IGF-1R in cancer cells is to employ anti-IGF-1R antibodies that bind to the extracellular domain of the receptor and inhibit its activation by ligands.The anti-tumor activity of some of these antibodies/i.e.IR3,1H7,EM164 and 19D12/has been described in tumor cell lines and xenografts in nude mice.CP-751,871 is a fully human monoclonal antibody that specifically binds to the IGF-1R.Clinical studies of CP-751,871 are now in progress.
