The "Shot-gun" method is one of the most popular tools in current proteomics study.In this method, digestion is the key process for protein profile via mass spectrometry (MS) analysis.Traditionally, digestion is often performed by free enzymes in solution, which is time-consuming, tedious and inconvenient for automation.In response to these challenges, an immobilized enzyme reactor in characteristic of rapid regeneration for high throughput digestion in proteomics was developed in this study.In detailed, a poly ((glycidyl methacrylate-co-acrylamide-co-methylenebisacrylamide) monolith was prepared in the 100 μm i.d.capillary.The monolith was initially coated with polyethylenimine polymer for adsorbing Cu2+, followed by chelating trypsin.
