Small interfering RNA(siRNA)is of considerable current interest in biology and medicine because it can elicit potent,target-specific knockdown of any mRNA,creating a useful and proven genetic surrogate tool. However,as with other antisense approaches, applying siRNA technology can prove challenging.Problems include enhancing siRNA stability, minimizing off-target effects, identifying sensitive sites in the target RNAs,and particularly ensuring efficient delivery.
