The general framework of promoter mechanism is dependent on protein-DNA interaction,in which different transcription factors (TFs) bind on the promoter region to enable its activity.An important step during transcription initiation is the open complex formation between RNAP and promoter sequence,which involves local separation of the two strands around the-35 and-10 regions.
