By engineering t-RNA and t-RNA synthetase, I have first genetically incorporated fluorescent amino acids in vivo.We are now using these environmentally sensitive fluorescent amino acids to probe enzyme mechanism, protein folding, and protein-protein interaction.Furthermore, because the fluorescent amino acids are pH sensitive, they are incorporated into cytokines to probe the kinetics of cytokine endocytosis process.
