晚期肾癌的治疗中,Mammalian Target of Rapamycin (mTOR)的靶向抑制剂有效果,并已经被FDA批准应用于临床.phosphatidylinositol 3-kinase (PI3-K)是调控mTOR的上游激酶,其抑制剂也在积极的临床试验中.针对其所可能产生的耐药性,我们研究了PI3K抑制剂LY 294002和Bcl-2抑制剂ABT-737针对肾癌细胞的协同作用.我们结果显示ABT-737可以显著增强PI3K抑制剂诱导肾癌细胞凋亡的作用,因而此类药物将可以显著增强PI3K抑制剂类药物在临床治疗肾癌的有效性,对临床治疗具有指导意义.申请人也对其分子机制做了研究,发现Mcl-1,XIAP,Bim的变化是其作用的分子基础；并最终导致caspase 3、caspase 8的激活.
