Ethyl (S)-4-chloro-3-hydroxybutyrate (ECHB) is an intermediate of Atorvastatin,a chiral drug for hypercholesterolemia.Rhodococcus erythropolis strain isolated from soil possessed the ability to convert 4-chloro-3-hydroxybutyronitrile into the corresponding acid.The activity was suggested to due to the successive actions of nitrile hydratase (NHase) and amidase.In this study,ammonium sulfate fractionation,DEAE ion exchange chromatography,and phenyl chromatography were used to purify NHase enzyme.By gel filtration chromatography and SDS-PAGE experiment,it was found to be a 92.3 kDa tetramer composed of two α and β subunits.The purified NHase had a high specific activity of 846 Umg-1 toward methacrylonitrile.NHase could be stabilized in the presence of organic acid such as butyrate,although it was thermolabile.The NHase exhibited a preference for aliphatic saturated nitriles over aliphatic unsaturated or aromatic ones.
