  NR2A and NR2B subunit-containing NMDA receptors have different dynamical properties and different cellular distribution in the cortex indicating that they may play different roles in cortical network activity, including gamma oscillations.In this study, gamma oscillations in frontal cortex EEG were assessed in rats before and after injection of non-selective (MK801, 0.2 mg/kg), NR2A-preferring (NVP-AAM077, 20 mg/kg), and NR2B-selective NMDA receptor antagonists (Ro25-6985, 10 mg), and vehicle.Results: MK-801 and NVP-AAM077 suppressed REM sleep for several hours and induced continuous enhanced gamma activity.When normal sleep-wake states returned, gamma activation was largest during REM sleep (46% and 100%), compared with other states (AW: 26%and 71%).Ro25-6985 had no effect on sleep-wake structure, but increased gamma power during REM sleep (37%,P=0.01 in the first and 23%, P=0.03 in the second 4 h period) with no change in other states (AW: 10 and 8%,P>0.3).Alteration of gamma oscillations due to subtype-specific changes in NMDA-R function may have serious implications for the mechanism and treatment of cognitive impairment.
