OBJECTIVE To explore the mechanism and signal pathway of IL-22 in the pathological damage organs of GVHD mice.METHOD The tissues sections of GVHD target organs were stained wi th HE for histopathologic detection and histopathologic score.The level of T-b et, RORγt, AHR were evaluated by PCR and RT-PCR.The expression of IL-22R and the infiltration of CD3+T cell in GVHD target organs were detected by immunohistochemistry.Western blot were conducted to detect the expression level of STAT3 , P-STAT3RESULTS: Compared with GVHD group, more CD3+ T cells infiltrated in impaired tissues of GVHD mice injected with IL-22.Meanwhile, transcription factor T-bet、 ROR γ t、AHR, respectively associated with Th1, Th17 and Th22 cells were elevated and the increased expression of IL-22R and its downstream protein P-STAT3 were detected in GVHD mice with IL-22 treated.
