MCAO大鼠局部脑血流的变化和缺血再灌注损伤程度的相关性研究

  稳定可靠的研究模型是取得真实实验结果的基础和关键。用线栓大鼠大脑中动脉(MCAO)法制作的实验性局灶性脑缺血/再灌注模型是被广泛采用的研究脑缺血病理生理过程和相关的脑缺血损伤的分子机制以及评价干预措施效应的比较理想的研究模型。但该模型也被认为由于多种原因导致其存在稳定性差,变异度大的缺点。有鉴于此,我们综合应用激光多普勒微循环检测仪的动态检测和神经行为学评分、脑梗死体积测定和模型大鼠24小时内死亡率等指标,研究了大脑中动脉线栓30 min、45 min和60 min所产生的局部脑血流的动态变化与其所致的缺血/再灌注损伤程度之间的关系。结果发现,模型变异度产生的直接原因主要是线栓后所产生的局部脑血流下降的程度及其血流波动幅度。通过控制局部脑血流的下降程度及其血流波动幅度,可以产生稳定的一致性高的时间依赖性的脑缺血/再灌注损伤后果。随着线栓大脑中动脉时间的延长,局部脑组织的低血流灌注时间延长,局部缺血组织的再灌注无复流现象持续加重,时间延长,脑缺血再灌注损伤程度加重。研究结果表明,局部脑组织血流灌注量下降的程度和低血流灌注持续的时间是缺血/再灌注损伤的关键性因素。严格控制局部脑血流的下降程度和缺血持续时间是产生稳定性高,一致性好的脑缺血再灌注损伤模型的重要举措。
