类水滑石材料作为新型纳米载体在植物细胞生物学中的应用

我们在此报道一种可负载生物活性材料进入活体植物细胞的新型纳米载体.类水滑石(Layered double hydroxides,LDHs)作为药物和生物分子载体被广泛应用在基于动物和人体细胞的细胞学研究中.由于植物细胞包裹着致密的细胞壁,而且其细胞膜具有与动物细胞不同的特性,LDHs在植物学中的应用仅有很少报道.为克服这一局限,我们合成了基于阳离子Mg2+Al3+和乳酸(lactate)的LDH材料,并通过在水相中的剥离而获得二维纳米片层.通过研究其对生物大分子的时间与浓度依赖的吸附,优化了其对FITC和DNA分子的吸附方法.再进一步通过细胞毒性试验表明,剥离的LDH对人体肾细胞系(293T)、烟草悬浮细胞系(BY-2)和拟南芥细胞具有很低的毒性.我们将293T,BY-2和拟南芥细胞与LDH-FITC和LDH-ssDNA-FITC偶联物共孵育发现剥离的LDH-lactate二维纳米片可作为有效低毒,而且应用简单的生物分子载体,将多种生物分子导入活体动植物细胞中.
