目的:探讨氟对成骨细胞的影响。方法:采用wister乳鼠颅盖骨成骨细胞原代培养的方法,观察不同剂量的氟化钠(NaF,10-7mol/L~10-3mol/L)对成骨细胞的细胞增殖、碱性磷酸酶(ALP)活性、培养液中骨钙素(BGP)含量、细胞蛋白质含量、转化生长因子β1基因(TGF-β1mRNA)表达和Ⅰ型胶原基因(COL-1mRNA)表达的影响。结果:低剂量NaF(10-7mol/L~10-5mol/L)可刺激细胞增殖,促进细胞ALP活性,增加细胞蛋白质合成(P<0.05);高剂量NaF(10-4mol/L、10-3mol/L)则抑制细胞增殖、降低细胞ALP活性,减少蛋白质合成(P<0.05)。各浓度NaF均促进细胞骨钙素分泌,与对照组相比有统计学意义(P<0.05)。各浓度NaF对TGF-β1mRNA的表达均无影响(P>0.05),但随浓度的改变TGF-β1mRNA的表达仍有变化趋势,即低剂量提高TGF-β1mRNA的表达,高剂量降低TGF-β1mRNA的表达,但无统计学意义。10-7mol/L、10-6mol/LNaF对COL-1mRNA的表达无影响(P>0.05),但当NaF为10-5mol/L时,COL-1mRNA的表达明显高于对照组(P<0.05);NaF为10-4mol/L、10-3mol/L时,COL-1mRNA的表达明显低于对照组(P<0.05),且有统计学意义。结论:NaF对成骨细胞增殖、早期分化及蛋白质合所抢医年斗大学医学币眨士掌位创势文成量呈双向调节,即小剂量呈现促进作用,大剂量为抑制作用。但对其晚期分化呈一致性,即促进作用。10一smol/L的NaF促进COL一lmRNA的表达,而过量氟则抑制COL一lmRNA的表达。关键词:氟化钠;成骨细胞;增殖;碱性磷酸酶;骨钙素;蛋白质合成;TGF一plmRNA;COL一lmRNAThe EffeCt of FIUoride on Osteoblasts ofNew Born RatS in ViroPostgraduate:Liu Feng SuPervisor:Liu kai一tai(DePartment of OeeuPational and Environmental Hygiene,sehool of Publie Health,Xinjiang Medieal University)AbstraCt objeetive:To study the effect of fluoride on osteoblasts.Methods:Osteoblasts were obtained from the ealvaria of 24一hnew born wister rat by using Primary eulture.The effeet ofNaF(一。一7mol/L一xo一3mol/L)on the eell proli化rati。n,the aetivityof alkaline PhosPhatase(ALP),Osteroealein,the eell Protein,theexPression level of TGF一p,mRNA and COL一1 mRNA ofosteoblasts were observed.Results:Low doses ofNaF(10一7mo一/L一10一smolzL)promoted prozireration,ALPaetivity and eontent of Protein of osteoblasts(P<0.05):whileh igh doses of NaF(一。一4mo一/L,10一3mol/L)deereased them(尸<
