Objective:AD is a form of brain damage to memory and cognitive function-based central nervous system degenerative disease symptom of behavioral and psychological abnormaties. Current ly not yet fully understand the causes of AD, Most of the drugs are in the treatment of clinical trials and the long-term use of such medicine easy to cause serious side effect.Therefore from the point of Chinese medical practitioners, to explore the effective treaatment has become the main trends in research. In recent years, clinical reports and experimental studies show that electro-puncture as an important component of Chinese medicine for the prevention and treatment of AD has a positive effect. Therefore, the application intracerebral injection of STZ,the establishment of rat model of AD, study of electro-puncture on learing and memory abilitities AD model rats. Effect of the study on learning and memory ability, hippocampus CaMKⅡ, NOS, BDNF expression,electro-puncture treatment of AD for the clinical application and to provide experimental basis for further research.Methods:Selective healthy Wistar male rats were 60. They were used for Morris test. We select 50 of them. Were randomly divided into normal group,moodel group, model control group,western medicine group and electro-puncture group. Methods of modeling micro-syringe on each side of intracerebraloventricular injection of 10% STZ 10 ul established AD model. Model control group were injected with normal saline instead of STZ.After modeling, electropuncture group after after electropuncture stimulation " Baihui", "Dazhui", "Shenshu", "Zusanli", "Taixi" points for treatment.With the mooris water maze test of learning and memory in rats, one randomly selected 50 ratrs were observed by immunohistoochemistry in hippocampus of rats CaMKⅡ、NO、BDNF expression.Results:(1) Behavioral:Compared with model group, the behavioral of the electropuncture group and weatern medicine group are significantly increased. Escape latency and swimming distance was significantly shorter (P<0.01).Rats in the former platform quadrant swim time, the percentage of the total swimming time was significantly higher. For the first time through the original platfprm in the model group, expression of CaMKⅡ、NO、BDNF was significant obvious in the model group. Compared with the model group, the expressions of electropuncture group and western medicine group were significantly increased. After the treatment, comparsion of the three group were significant difference (P<0.01).Conclusion:(1)Electro-puncture treatment AD rats significantly improved the memory and learning abilities. (2) Electro-puncture treatment can improve CaMKⅡ、NO、BDNF expression in the AD rat brain
