苹果MdHIRs蛋白与MdJAZs蛋白互作对花青苷积累的调控

本文以红星和富士(Malus×Domestica Borkh)果实为材料,利用瞬时表达技术,鉴定了MdHIR2、MdHIR4和MdJAZ2在花青苷合成调控中的的功能。主要结果如下:1.MdDAZ2基因抑制苹果果皮的花青苷积累为了研究MdJAZ2基因对苹果果皮着色的影响,构建了35S::MdJAZ2瞬时表达载体,注射进红星果实果皮中,结果发现MdJAZ2能有效抑制苹果果皮花青苷的积累,并且花青苷合成途径中的相关基因的表达受到明显抑制。说明MdJAZ2参与了苹果果皮花青苷的积累。2.苹果MdJAZs蛋白与MdHIRs蛋白相互作用为了更深入的探求MdJAZ2对苹果果皮花青苷积累调控的机制,利用酵母筛库的实验技术筛选到MdHIR2蛋白,酵母双杂交及Pull-Down实验技术进一步证明了MdJAZs蛋白与MdHIRs蛋白的相互作用。3.MdHIRs基因抑制苹果果皮中花青苷的积累蛋白MdHIRs可以与MdJAZs相互作用,本文同样用注射的方法对MdHIRs基因在果皮中花青苷的积累中的功能进行了验证,结果发现在苹果果皮中过表达MdHIR,能显著抑制花青苷的积累,而沉默MdHIR基因,苹果果皮中的花青苷积累增多。
