目的:观察祛痰通络汤(QTTLD)对糖尿病肾病(DN)大鼠足细胞裂孔隔膜蛋白分子(podocin)表达的影响,探讨其防治DN的作用机制。方法:制作大鼠糖尿病肾病模型,分为6组:正常对照组、模型对照组、贝那普利治疗组、祛痰通络汤低剂量组、祛痰通络汤中剂量组、祛痰通络汤高剂量组,各组分别干预8周。8周后观察各组24 h尿蛋白定量,观察肾脏病理学变化,并采用免疫组化观察肾组织podocin蛋白表达的变化。结果:经过祛痰通络汤治疗后,24 h尿蛋白定量下降(P<0.05);病理改变减轻;podocin蛋白表达增加(P<0.05)。结论:祛痰通络汤对DN大鼠具有一定的肾脏保护作用,其机制可能与恢复肾组织足细胞podocin表达有关。
Objective: To observe the effects of Qutan Tongluo Decoction (QTTLD) on the expression of podocin in podocytes of diabetic rats with nephropathy (DN), and to explore its mechanism of action in preventing and treating DN. Methods: The rat model of diabetic nephropathy was made and divided into 6 groups: normal control group, model control group, benazepril treatment group, low dose of Qutan Tongluo Decoction group, middle dose of Qutan Tongluo Decoction group, Treating high-dose group, each group were intervention for 8 weeks. After 8 weeks, the urinary protein excretion of 24 hours was observed, the pathological changes of kidney were observed, and the expression of podocin protein in renal tissue was observed by immunohistochemistry. Results: After expelling Phlegm Tongluo decoction, 24 h urinary protein was decreased (P <0.05), pathological changes were alleviated and podocin protein expression was increased (P <0.05). Conclusion: Qutan Tongluo decoction has a protective effect on kidney of DN rats, and its mechanism may be related to the podocin expression in parenchymal cells of kidney.
