目的:检测转染吲哚胺2,3-双加氧酶(IDO)的MFC胃癌细胞移植瘤中E-cadherin、N-cadherin及IL-6的表达水平。方法:建立MFC胃癌细胞移植瘤模型,分为MFC胃癌细胞组、pcDNA3.1-MFC细胞(空质粒)组、pcDNA3.1-IDO-MFC细胞组,免疫组化方法检测3组的E-cadherin和N-cadherin表达情况,酶联免疫吸附试验检测3组荷瘤小鼠血清液中IL-6的表达水平。结果:pcDNA3.1-IDO-MFC组移植瘤组织中,E-cadherin的表达较空质粒组和MFC细胞组明显减少(P<0.05),N-cadherin的表达较其他两组明显增多(P<0.05)。pcDNA3.1-IDO-MFC细胞组荷瘤小鼠血清IL-6水平较其他两组明显增高(P<0.05)。结论:IDO转染导致胃癌细胞移植瘤中E-cadherin水平下调、N-cadherin和IL-6水平上调,该结果为IDO参与肿瘤细胞的侵袭和转移提供实验依据。
OBJECTIVE: To detect the expression of E-cadherin, N-cadherin and IL-6 in transplanted MFC gastric cancer cells with indoleamine 2,3-dioxygenase (IDO). Methods: The model of gastric cancer cell xenografts in MFC was established and divided into 3 groups: MFC gastric cancer cell group, pcDNA3.1-MFC cell (empty plasmid) group, pcDNA3.1-IDO-MFC cell group, immunohistochemical method to detect E-cadherin And N-cadherin expression. ELISA was used to detect the level of IL-6 in serum of three tumor-bearing mice. Results: The expression of E-cadherin in pcDNA3.1-IDO-MFC group was significantly lower than that in empty plasmid group and MFC cell group (P <0.05), and the expression of N-cadherin was significantly increased in the pcDNA3.1-IDO-MFC group compared with the other two groups 0.05). The level of IL-6 in pcDNA3.1-IDO-MFC cells group was significantly higher than that in other two groups (P <0.05). CONCLUSION: IDO transfection results in downregulation of E-cadherin and up-regulation of N-cadherin and IL-6 in gastric cancer xenografts. This result provides experimental evidence for the involvement of IDO in tumor cell invasion and metastasis.
